Urolithin A is a metabolite formed when certain gut bacteria convert plant compounds known as ellagitannins and ellagic acid.

The process can be simplified as:

No.

There is currently no established Recommended Dietary Allowance, or RDA, for Urolithin A.

That makes Urolithin A different from established essential nutrients such as vitamin C, calcium, or vitamin D.

What Urolithin A Doses Have Actually Been Studied?

Early Human Research: 10–1,000 mg

A first-in-human study published in Nature Metabolism evaluated the safety, pharmacokinetics, and biological activity of Urolithin A in older adults.

The research included multiple doses, ranging from relatively low amounts to doses as high as 1,000 mg.

Researchers found that orally administered Urolithin A was bioavailable and that repeated supplementation produced changes in several molecular signatures associated with mitochondrial and cellular metabolism.

The study provided important early evidence that Urolithin A could be administered orally to humans.

What Does the Research Say About 500 mg of Urolithin A?

A 2022 randomized, double-blind, placebo-controlled trial published in Cell Reports Medicine studied Urolithin A in overweight, untrained middle-aged adults.

A total of 88 participants were randomized to receive:

  • Placebo
  • 500 mg Urolithin A per day
  • 1,000 mg Urolithin A per day

for four months.

Researchers reported statistically significant improvements in certain measures of hamstring muscle strength in both the 500 mg and 1,000 mg groups compared with placebo.

However, other outcomes did not demonstrate clear significant differences.

For example, improvements in some quadriceps measures and hand-grip strength were not statistically significant between groups.

This is important because the results do not justify reducing the study to a claim such as:

“500 mg has been proven to improve all aspects of muscle performance.”

What About 1,000 mg of Urolithin A?

The same trial also investigated 1,000 mg per day.

Participants receiving 1,000 mg demonstrated improvements in some strength and exercise-related measurements.

But this does not establish that 1,000 mg is universally better than 500 mg.

In fact, the relationship between dose and outcome was not consistently linear across every endpoint.

Some outcomes improved at both doses, some showed stronger numerical changes at one dose, and others were not significantly different from placebo.

Therefore:

1,000 mg should not automatically be interpreted as “twice as effective” as 500 mg.

The review found effects on some mitochondrial, inflammatory, strength, and endurance-related outcomes, but also reported outcomes where no significant effects were found.

The authors concluded that additional research across more physiological systems and over longer intervention periods is needed.

What Does the 2026 Meta-Analysis Add?

A newer systematic review and meta-analysis published in 2026 examined randomized controlled trials of Urolithin A and muscle-related outcomes.

It identified five randomized controlled trials involving 236 participants.

The authors emphasized several limitations:

  • Small study populations
  • Short study durations
  • Different participant groups
  • Different doses
  • Different outcome measurements
  • Limited ability to combine results statistically

For the 6-minute walk test, the pooled improvement favored Urolithin A numerically, but the result was not statistically conclusive, and the certainty of evidence was rated low.

Other findings related to strength, endurance, aerobic capacity, and mitochondrial biomarkers were described as exploratory.

This is an important update.

It means the responsible conclusion in 2026 is not:

“Urolithin A is clinically proven at X mg.”

It is:
